This Uncharted 4: A Thief's End speedrun walkthrough shows the complete game from start to finish (cutscenes are not included).
To earn the /"Charted! – Speedrun/" trophy you have to complete the game in 6 hours or less. I managed it in 4 hours, 42 minutes and 34 seconds.
The most important thing to remember is that cutscenes add to your overall time, so you have to watch them all. Some can't be skipped.

I also ended up with an accuracy of 83%. To get the Sharpshooter trophy you need 70% accuracy.

Tips for speedruns:
– Cutscenes extend your playing time! You must skip them!
– The timer stops during loading screens and when you pause the game.
– Play on Explorer difficulty (trophies will not be disabled)
– Enable automatic aiming in the options (this will not disable trophies)
– Enable Infinite Ammo and use the Barok .44 revolver from the weapon selector, it kills almost anything in one hit. Cheats are unlocked after you win the game for the first time on any difficulty (cheats do not disable trophies).
– Remember you can fire from the hip while running past enemies. Works great at close range.
– You don't have to kill every single enemy. Sometimes you can run past entire enemy encounters without killing anyone.
– You are allowed to create manual saves and exit the application. It is not necessary to do this in one session, the trophy will still be unlocked.
– If you encounter a progress error, just exit the application and continue the story and it should load fine (endless loading screen, game crash, NPCs not doing what they're supposed to).
– When you die or restart checkpoints, your time continues to count (the time until the last checkpoint is not reset).
– You can always check your playing time in the stats. Compare it to the time in the video to get an idea of where you stand. I still had plenty of time left for the cup.
– The puzzles can take a lot of time if you don't do them perfectly. Check them out in the video, I'll show them as quickly as possible.

All in all, with auto-aim and cheats, it's not too difficult to meet the 6 hour requirement, just remember to skip all the cutscenes.
